Specificity

Clone 1E11 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Nucleoporin SEH1L. It targets an epitope within 21 amino acids from the C-terminal region.

Immunogen

KLH-conjugate linear peptide corresponding to 21 amino acids from the C-terminal region of human Nucleoporin SEH1, isoform A.

Application

Quality Control Testing

Evaluated by Western Blotting in PC3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SEH1L in PC3 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SEH1L in MDA-MB-231 and A549 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound SEH1L with a KD of 2.4 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SEH1L in A549 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SEH1L in human cerebral cortex tissue sections.

Target description

Nucleoporin SEH1 (UniProt: Q96EE3; also known as GATOR complex protein SEH1, Nup107-160 subcomplex subunit SEH1, SEC13-like protein, SEH1L) is encoded by the SEH1L (also known as SEC13L, SEH1) gene (Gene ID: 81929) in human. Nuclear pore complexes (NPCs) are composed of multiple copies of approximately 30 different proteins termed as nucleoporins. In vertebrates, one such building block of the NPC, the Nup107-160 complex, contains six different nucleoporins. SEH1L is one of the components of the Nup107-160 subcomplex. This subcomplex is required for normal kinetochore microtubule attachment, mitotic progression, and chromosome segregation. The SEH1L subunit that contains six WD domains appears to be only weakly associated with the Nup107-160 subcomplex. Depletion of Nup107-160 complex or depletion of SEH1L is shown to induce mitotic delay. SEH1L depleted cells also display impaired chromosome congression, reduced kinetochore tension, defects in and kinetochore microtubule attachments. SEH1L is also a component of the GATOR subcomplex 2 and functions within the amino acid-sensing branch of TORC1 signaling pathway. It is reported to indirectly activate mTORC1 signaling pathway through the inhibition of the GATOR 1 subcomplex. Knockdown of SEH1L is reported to inhibit mTORC1 activity. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
