General description

Sphingosine kinase 2 (Sphk2), along with its sister enzyme Sphk1, is an important kinase needed to form the important intra and extracellular signaling lipid mediator and neuroprotective factor SPP/S1P (sphingosine 1-phosphate). Sphk2 is important in blood vessel development, brain development, as well as an anti-apoptotic factor, and it mediates many proliferation mediated responses such as those surrounding insulin and its mitogenic ability. Sphk2 is localized to the lysosome membrane primarily but can also be found in the cytoplasm. Interestingly, recent research indicates that ApoE regulates the secretion Sphingosine 1-phosphate in the brain, and SPP/S1P in turn positively regulates glutamate secretion and synaptic strength in hippocampal neurons, thus Sphk1/2 plays an important role in synaptic plasticity and neuronal activity connectivity in the brain as well.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Sphk2 in 10 µg of mouse lung tissue lysate.

Quality

Evaluated by Western Blotting in RAW264.7 cell lysate.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
