Description générale

Insulin receptor substrate-1 (IRS-1) is among the four IRS proteins (IRS-1-IRS-4) that mediate signalling by insulin receptor (IR) and insulin-like growth factor (IGF) receptor. IRS-1 has a pleckstrin homology domain and a phosphotyrosine-binding domain. Phosphorylation of the Tyr1179 residue in the SH-1 negatively regulates insulin. IRS-1 has the abililty to translocate to the nucleus, the mechanism of which is not very clear. Estrogen receptors (ER) α and β have a strong affinity to IRS-1, an important interaction in breast cancers and medulloblastomas. [1] The expression of IRS-1 in prostate cancer is associated with lower tumor cell invasiveness. Along with ER α, IRS-1 also modulates transcriptional activity involved in the development of breast and prostate cancer. [1] The signalling pathway involving IGF-IR-IRS-1 in cells expressing JCV T-antigen has been reported to affect the proliferation, survival and DNA repair in the cells. Phosphorylation of IRS-1 at Ser312 and Ser616 results in the negative regulation of insulin signalling in humans
Anti-phospho-IRS-1 (pSer312) specifically recognizes human IRS-1 (pSer312). It may react with mouse and rat (100% homology) IRS-1.

Immunogène

synthetic phosphopeptide derived from the region of IRS-1 that contains serine 312. This sequence is 100% homologous with mouse and rat IRS-1.

Application

Anti-phospho-IRS-1 (pSer312) antibody may be used for detection by immunoblotting, in stimulated CHO cell line expressing human insulin receptor, at a working dilution of 1:1000.

Forme physique

Solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), pH 7.3 containing 50% glycerol, 1.0 mg/mL BSA (IgG, protease free) and 0.05% sodium azide.
