Immunogène

synthetic peptide corresponding to amino acids 205-221 located in a region near the C-terminus of Arabidopsis AtEB1b, conjugated to KLH. This sequence is highly conserved (single amino acid substitution) in AtEB1a and AtEB1c.

Application

Anti-AtEB1b (C-terminal) antibody produced in rabbit is suitable for western blotting at a concentration of 1-2μg/mL using leaves extract (cytosolic fraction) of transgenic Arabidopsis seedlings expressing YFP-tagged AtEB1b.

Actions biochimiques/physiologiques

End-binding 1 (EB1) proteins are plus-end-tracking proteins that are specifically localized to growing microtubule plus ends that regulate microtubule and interact with intracellular targets. The plant-specific EB1 subtype functions on the spindle microtubules by accumulating in the prophase nucleus. The microtubule-end-binding protein expressed in Arabidopsis thaliana cells (is called as AtEB1a) is fused with green fluorescent protein (GFP) resulting in labelling of spindle poles. During interphase, AtEB1a-GFP labels the microtubule plus end and also marks the minus end. The microtubule associated protein END BINDING 1b (EB1b) is involved as mutants and dampens root responses to touch and enhances gravitropism. In animals, EB1 proteins have acidic C-terminal tails that might induce an auto-inhibitory conformation.
