The S100 protein 330M-18 consists of both alpha and beta subunits. This protein belongs to the family of calcium-binding proteins, similar to calmodulin and troponin C. Specifically, S100A is composed of an alpha and beta chain, while S100B is composed of two beta chains. Furthermore, the S100 protein is expressed in antigen-presenting cells, including Langerhans cells in the skin and interdigitating reticulum cells in the paracortex of lymph nodes.

S-100-Protein wird in normalen Melanozyten, Langerhans-Zellen, Histiozyten, Chondrozyten, Lipozyten, Zellen der Skelett- und Herzmuskulatur, Schwann-Zellen, epithelialen und myoepithelialen Zellen der Brust, Speichel- und Schweißdrüsen sowie in Gliazellen gefunden.[1][2][3]Von diesen Zellen abstammende Neoplasien exprimieren ebenfalls das S-100-Protein, wenn auch nicht gleichmäßig.[1][2][4][5] Eine große Anzahl gut differenzierter Tumore von Speicheldrüsen, Fett- und Knorpelgewebe[4] sowie von Schwann-Zellen abgeleitete Tumore exprimieren das S-100-Protein. Beinahe alle malignen Melanome und Fälle von Histiozytose X erbringen ein positives Ergebnis für S-100-Protein.[5][6] Trotz der Allgegenwart des S-100-Proteins ist sein Nachweis sehr wertvoll für die Identifizierung verschiedener Neoplasien, insbesondere von Melanomen.[1][2][4][5][6][3]
