General description

Folin & Ciocalteus Phenolreagens wird am häufigsten im Lowry-Test zum Bestimmen der Proteinkonzentration eingesetzt.[1][2] Es wird auch zur Quantifizierung der Gesamtphenole eingesetzt. In diesem Test wird Protein mit Kupfer(II) in einem modifizierten Biuretreagens (alkalische Kupferlösung,
stabilisiert mit Natriumkaliumtartrat) vorbehandelt. Die Zugabe von Phenolreagens erzeugt Chromogene, die eine steigende Absorption von 550 bis 750 nm bereitstellen. Normalerweise wird die Absorption an Peak (750 nm) oder Schulter (660 nm) verwendet, um eine Proteinkonzentration von 1 bis 100 mg/ml zu quantifizieren, während eine Absorption bei 550 nm verwendet wird, um höhere Proteinkonzentrationen zu quantifizieren.
In Abwesenheit von Kupfer wird die Farbintensität primär durch den Tyrosin- und Tryptophangehalt des Proteins bestimmt, und in geringerem Maße durch Cystein und Histidin. Kupfer(II) hat keine Auswirkung auf die Farbbildung durch Tyrosin, Tryptophan oder Histidin, verringert aber die Farbbildung durch Cystein.[1][3][4][5][6]
Viele Modifikationen des ursprünglichen Assay-Verfahrens wurden publiziert,[3] einschließlich Verfahren zum Verbessern der Farbentwicklung,[5][7] zum Bestimmen des Gehalts von unlöslichen Proteinen[1][8] und zum Automatisieren des Verfahrens.[9] Verbindungen, die viele Puffer, Chelatbildner, Detergenzien und cyclische organische Verbindungen einschließen können den Lowry-Protein-Assay beeinträchtigen.[3]
Folin & Ciocalteus Phenolreagens kann auch als Sprühreagens bei chromatografischen Verfahren eingesetzt werden.

    1. This product is not tested according the BP or JP guidelines. This reagent is not recommended for the determination of phenols. HIstorically, the performance of this product in the determination of phenolic compounds, typically run in wineries, has proven to be unpredictable. It is recommended for use primarily in Lowry protein determination.

    1. The type and amount of non-phenolic compounds present is a given sample will determine the impact on the total phenol values. The use of a standard curve is critical. Factors such as pH adjustment can minimize the effects. Reducing compounds may be identified and quantified separately or removed from the sample by various means.
