Beschreibung

General description

Ein zellpermeabler, reversibler, substratkompetitiver und potenter Inhibitor der Autophosphorylierung der epidermalen Wachstumsfaktor-Rezeptor-Kinase (IC₅₀ = 100 nM). Die Hemmung von JAK2 durch AG 490 blockiert selektiv das Wachstum leukämischer Zellen in vitro und in vivo, indem sie den programmierten Zelltod auslöst, ohne schädliche Auswirkungen auf die normale Hämatopoese zu haben. Hemmt die konstitutive Aktivierung der STAT-3-DNA-Bindung und das IL-2-induzierte Wachstum von MF-Tumorzellen. AG 490 hemmt nachweislich auch die Autokinase-Aktivität von JAK3. Eine 100-mM-Lösung (5 mg/170 µl) von AG490 (Best.-Nr. 658411) in DMSO ist ebenfalls erhältlich.
