General description
Eine zellpermeable Nitroindolguanidinhydrazon-Verbindung, die als potenter, ATP-kompetitiver und hochgradig selektiver Chk2-Inhibitor dient (IC50 = 138 und 570 nmol/l gegen die Phosphorylierung von Histon H1 durch Chk2 in Gegenwart von 10 µmol/l bzw. 1 mmol/l ATP), jedoch eine deutlich geringere oder keine Wirkung gegen einen Panel aus 52 anderen zellulären Kinasen, einschließlich Chk1 (IC50 = 15,73 und 55 µmol/l gegen Substratphosphorylierung bzw. Chk1-Autophosphorylierung mit 10 µmol/l ATP) aufweist. Unterdrückt die durch Strahlung eingeleitete Phosphorylierung von Chk2 (an Ser516) und sein Downstream-Substrat Cdc25c (an Ser216) in MCF7-Kulturen (IC50 <10 µmol/l) und verringert die Sensitivität von murinen Wildtyp-Thymozyten gegenüber durch Strahlung eingeleiteter Apoptose auf das in Chk2-/--Kulturen gemessene Maß.
