Cell Line Description

The cell line TRA-171 has been established from freshly hatched larvae of Toxorhynchites amboinensis. After eggs had been surface-sterilised, they were allowed to hatch in MM / VP12 medium and were mechanically damaged at the anterior or posterior ends of the body in 0.25% trypsin. After washing and cultivation at 22 - 25°C small patches of cells were visible leading to the isolation of the cell line TRA-171 after 72 days in culture. The cells have been adapted to grow in a 1:1 mixture of Leibovitz L-15 and Mitsuhashi and Maramorosch media plus supplements. Four serotypes of laboratory adapted and three type strains (1, 2 and 3) of un-adapted dengue virus will replicate in the cells at 28°C.

Culture Medium

L15: Mitsuhashi and Maramorosch Insect Medium (1:1) + 2mM Glutamine + 0.05% BSA + 1% NEAA + 10% Heat Inactivated Foetal Bovine Serum (HI-FBS)

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 to 1:4 i.e. seeding at 2-4 x 10,000 cells/cm2 using 0.25% trypsin for 5 minutes only at 28°C, after quenching the trypsin with fresh medium dislodge cells by gentle scraping;  CO2 not required; 28°C. Fr
