General description

ReadyTube® is ready to use media to provide the highest level of quality and testing confidence for improving accuracy and significantly reducing the risk of false results. ReadyTube® modified semi-Solid RAPPAPORT-VASSILIADIS medium is used for the selective enrichment and presumptive detection of Salmonella sp. This medium is a semi-solid modification of Rappaport-Vassiliadis enrichment broth and is used for motility enrichment in Petri dishes. It is a rapid and sensitive medium for the isolation of Salmonella sp. and can be seeded directly after pre-enrichment.

Application

ReadyTube® modified semi-Solid RAPPAPORT-VASSILIADIS medium is used for the selective enrichment and presumptive detection of Salmonella from food, animal feed, and environmental samples from primary production stage

Analysis Note

Peptone and casein hydrolysate provide nitrogenous, carbonaceous, B complex vitamins and other essential growth nutrients. Phosphate provides buffering capacity to the medium. Malachite green serves as a selective agent. The addition of novobiocin as a supplement in the medium selectively inhibits most Gram-positive organisms. Sodium chloride maintains the osmotic equilibrium of the media. Salmonella generally survives a slightly high osmotic pressure (due to MgCl2 in the medium), grows at a slightly low pH, and is resistant to malachite green compared to other bacteria. Its efficiency is due to the ability of Salmonellae to migrate into the semi-solid medium, thus forming opaque halos of growth. All these factors make the medium selective for the isolation of Salmonella. Also, the semi-solid nature of the medium helps to differentiate the motile Salmonella from non-motile ones.
