General description

The protein C receptor (PROCR) gene, spanning 6kb with 4 exons, is mapped to human chromosome 20q11.22. The gene encodes a receptor for activated protein C/ endothelial protein C receptor (EPCR). PROCR is a 46kDa, N-glycosylated type I membrane protein and is expressed on endothelial cells. The encoded protein contains 221 amino acids and is characterized by an extracellular domain, a transmembrane domain and a 3 amino acid intracytoplasmic sequence.

Application

Biochem/physiol Actions

Protein C receptor (PROCR)/ endothelial protein C receptor (EPCR) plays a vital role in factor FVII/FVIIa-mediated blood clotting and inflammation. The encoded protein also plays a crucial role in protein C (PC) anticoagulant pathway. In addition to its role in coagulation, PROCR is also implicated in the inflammation control pathway. Mutation in the gene increases the risk of susceptibility to both arterial and venous thrombotic disease. Additionally, variation in the gene is also associated with the severe malarial infection.
