General description

Anti-eukaryotic translation initiation factor 2C 3 (EIF2C3)/protein argonaute-3 (Ago3) is developed in rabbit using as immunogen a synthetic peptide corresponding to human EIF2C3/Ago3 conjugated to KLH. Argonaute proteins (molecular mass 100 kDa) are characterized by piwi-argonaute-zwille (PAZ) and PIWI domains. Ago subfamily consists of four members, hsAgo1-4. hsAgo2 and Ago3 associate primarily with miRNA. Ago proteins localize to the cytoplasm of somatic cells and are concentrated in cytoplasmic processing bodies. A member of this group, Ago1, is associated with the Golgi and endoplasmic reticulum. The gene is located on chromosome 1 in a cluster of closely related family members including Ago 1 and Ago 4.
Argonaute 3, RISC catalytic component (Ago3), also known as eukaryotic translation initiation factor 2C, 3 (EIF2C3), is encoded by the gene mapped to human chromosome 1p34.3. The encoded protein belongs to the argonaute family of proteins and is characterized with a PAZ (Piwi/Argonaute/Zwille) domain and PIWI domains.

Immunogen

synthetic peptide corresponding to amino acids 13-25 of human EIF2C3/Ago3 conjugated to KLH. The corresponding sequence differs by one or two amino acid in rat and mouse, respectively.

Biochem/physiol Actions

Argonaute proteins play a vital role in RNA interference (RNAi). These proteins are also involved in germline development, stem cell fate determination, as well as epigenetic regulation and transposon silencing. Ago3 facilitates the regulation of let-7a-3p expression. Mutation in the gene might lead to the development of Wilms′ tumors.
hAgo2 has been shown to have slicing activity. This region is lost in Wilms′ tumors, which are hypothesized to be caused by defects in embryonic kidney development, which disturb the capacity of metanephrogenic precursor cells to differentiate.

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
