General description

Reagent-cartridge VHL-PEG2 is a reagent cartridge for the Synple 2 Automated Synthesizer that enables the synthesis of molecules for targeted protein degradation and PROTAC® (proteolysis-targeting chimeras) research. The cartridge contains all necessary reagents for linking the user selected aldehyde or ketone to the amine terminated linker through a reductive amination. This cartridge features a von Hippel-Lindau (VHL) recruiting ligand ((S,R,S)-AHPC) and PEG linker. Because even slight alterations in ligands and crosslinkers can affect ternary complex formation between the target, E3 ligase, and degrader, many analogs are prepared to screen for optimal target degradation.

Scale: up to 0.1 mmol
Linker: PEG2
E3 ligase target and ligand: von Hippel-Lindau (VHL), (S,R,S)-AHPC
Terminal functional group: amine
Compatible substrates: ketone, aldehyde

Cartridge contents:
  • Reducing agent (supported cyanoborohydride)
  • Base (supported triethylamine)
  • SCX (for purification)
  • Degrader building block linker reagent
