General description

RAD51 (previously known as DNA repair protein RAD51 homolog 1) is a member of the Rec A family involved in DNA repair. RAD51 interacts with BRCA1 and BRCA2, which are both involved with breast cancer, to activate homologous recombination and DNA damage repair. BRCA1 appears to be present in heredity forms of breast cancer that results in the transition of G to A on codon 150, thereby converting ARG to GLN. BRCA2 regulates intercellular localization and DNA binding ability. Loss of BRCA2 results in genomic instability and tumorgenesis. RAD51 acts as ATPase to repair double strand breaks and also unwinds DNA to form a helical nucleoprotein filament. RAD51 is predominantly expressed in the testis and thymus but can also be found in the small intestine, placenta, colon, pancreas and ovaries.

Specificity

This antibody recognizes RAD51.

Physical form

Affinity purified
Purified rabbit polyclonal antibody precipitated in a solution of 50% saturated ammonium sulfate and PBS containing no preservatives.

Storage and Stability

Stable for 1 year at 2-8ºC from date of receipt. To reconstitute the antibody, centrifuge the antibody at a moderate speed (5,000 rpm) for 5 minutes. Carefully remove the ammonium sulfate/PBS buffer solution and discard; 10μL of residual ammonium sulfate solution will not affect the re-suspension of the antibody. Do not let the protein pellet dry, as severe loss of antibody reactivity can occur. Re-suspend the antibody pellet in 100L either standard PBS or TBS (pH 7.3-7.5). DO NOT VORTEX. Mix by gentle stirring with a wide pipet tip or gentle finger-tapping. Let the precipitated antibody rehydrate for 1 hour at 4-25°C prior to use. Small particles of precipitated antibody that fail to re-suspend are normal. Vials are overfilled to compensate for any losses.

Cell cycle regulation and DNA repair following damage are essential for maintaining genome integrity. DNA damage activates checkpoints in order to repair damaged DNA prior to exit to the next phase of cell cycle. Recently, we have shown the role

The role and therapeutic promise of poly-ADP ribose polymerase (PARP) inhibitors in anticancer chemotherapy are increasingly being explored, particularly in adjuvant or maintenance therapy, considering their low efficacy as monotherapy agents and their potentiating effects on concurrently administered contemporary chemotherapeutics.
