Biochem/physiol Actions

General transcription factor for RNA Polymerase II.
The amino terminal region of TFIIB contains a zinc ribbon fold composed of antiparallel Β strands. Mutational studies show that this domain is required for recruitment of RNA Pol II for transcriptional initiation. [1]

Unit Definition

One gel shift unit is the amount of TFIIB required to form 0.2 pmol of TFIIB/TFIID/DNA complex as detected by gel shift.

Physical form

Solution in 20 mM Tris-HCl, pH 7.4, 100 mM NaCl, 2 mM DTT, 1 mM EDTA, 0.1% Triton® X-100.

The function of the conserved zinc-binding domains in the related Pol II- and Pol III-specific factors TFIIB and Brf was investigated. Three-dimensional structure modeling and mutagenesis studies indicated that for both factors, the functional surface of the zinc ribbon fold
