Specificity

Recognition sequence: 5′-G/CTAGC-3′
Cutting results: a 2-10-fold Nhe I overdigestion of 1 μg λ DNA substrate results in 100% cutting.
Heat inactivation: Inactivated at 65 °C for 20 minutes.

Application

NheI is a restriction endonuclease that is used in molecular biology to cleave DNA at the recognition site 5′-G/CTAGC-3′ to yield fragments with 5′-cohesive termini compatible with ends generated by AvrII, SpeI and XbaI.

Supplied with 10x Restriction Enzyme Buffer SM (B3158).
Comment: High salt concentrations will inhibit activity of Nhe I.
